I think it's too hard to post just one. There's so many great movies which are best in each of their genres. But I fully agree that The Holy Grail is awesome! One of my favies.
Other favourites would be:

Gladiator
Ace Ventura movies
Batman: The Dark Knight (And the first movie for that matter)
